Estresse oxidativo e alterações morfológicas no hipocampo da prole de ratas hiperglicêmicas

O diabetes mellitus gestacional expõe o feto a um ambiente hiperglicêmico capaz de comprometer o desenvolvimento do sistema nervoso central, especialmente do hipocampo, estrutura altamente suscetível ao estresse oxidativo e às alterações morfológicas. O objetivo deste estudo foi avaliar as alterações morfológicas e os marcadores de estresse oxidativo no hipocampo da prole de ratas expostas à hiperglicemia durante a gestação. Ratas Wistar tiveram hiperglicemia induzida por estreptozotocina no quinto dia de gestação. Os hipocampos da prole foram coletados aos 7, 30 e 90 dias de vida para análises morfológicas e bioquímicas, incluindo a avaliação do equilíbrio redox. Os dados foram submetidos à análise estatística apropriada. As ratas hiperglicêmicas apresentaram hiperglicemia persistente durante a gestação, sem diferença expressiva na evolução da massa corporal em relação ao grupo controle. A prole exibiu menor índice de massa corporal, alterações morfológicas persistentes nos subcampos do hipocampo em todas as idades avaliadas, aumento da produção de espécies reativas de oxigênio, redução da capacidade antioxidante total e diminuição da atividade da superóxido dismutase. Além disso, observou-se redução da ativação da caspase-3 aos 30 e 90 dias de idade. Conclui-se que a hiperglicemia materna promove alterações morfológicas persistentes e comprometimento da capacidade antioxidante do hipocampo da prole, evidenciando que a exposição intrauterina ao ambiente hiperglicêmico interfere no desenvolvimento neural e mantém repercussões até a idade adulta em ratos.
